Rocket Pharmaceuticals, Inc. (NASDAQ:RCKT) on Monday shared a clinical safety update concerning the first three patients treated in its global, pivotal Phase 2 trial for Danon disease.
Danon disease is a rare, inherited genetic condition characterized by cardiomyopathy (weakening of the heart muscle), skeletal myopathy (muscle weakness) and intellectual disability.
The patients received RP-A501 under a recently modified protocol, demonstrating promising early safety results that pave the way for further clinical progress.
Promising Early Safety Under Modified Protocol
The commercial-stage biotechnology firm, which specializes in advancing genetic medicines for rare and inherited cardiovascular disorders, administered a recalibrated RP-A501 dose of 3.8 × 10¹³ GC/kg to the trial participants.
The revised dosing was carefully paired with a newly refined immunomodulatory regimen consisting of corticosteroids, rituximab and sirolimus.
The company administered the treatments sequentially, maintaining a minimum spacing of four weeks between each patient’s infusion. Thus far, the modified clinical approach has yielded highly positive safety outcomes.
Researchers have observed no occurrences of capillary leak syndrome, thrombotic microangiopathy (TMA), or any other significant safety concerns among the initial group.
FDA Engagement and Trial Progression
The pivotal Phase 2 clinical trial was originally designed as a single-arm study intended to evaluate a total of 12 patients.
Following the successful initial safety data from this early cohort, the drug developer is currently engaged in active discussions with the U.S. Food and Drug Administration (FDA).
These critical regulatory talks aim to secure alignment on the appropriate pathway to dose the remaining trial participants and bring the pivotal study to a successful completion.
The company expects to release another update regarding its regulatory progress with the FDA during the second half of 2026.
