Enliven Therapeutics Inc. (NASDAQ:ELVN) drew an Outperform rating from William Blair on Tuesday as the firm initiated coverage, citing the potential for lead drug candidate ELVN-001 to reshape treatment of chronic myeloid leukemia (CML).
ELVN-001 Shows Positive Phase 1 Data
Enliven Therapeutics’ lead drug, ELVN-001, is a small-molecule kinase inhibitor designed to specifically target the BCR::ABL1 gene fusion, the oncogenic driver of CML.
In June, the company announced positive data from the ongoing ENABLE Phase 1 clinical trial evaluating ELVN-001 in patients with previously treated CML.
The data showed 61% overall major molecular response (MMR) and 48% MMR by 24 weeks in the 80 mg once daily (QD) Phase 1b cohort.
Response rates were higher in earlier-line patients, with 67% overall MMR and 55% achieving MMR by 24 weeks among patients with only one or two prior unique tyrosine kinase inhibitors (TKIs).
Enliven Therapeutics Has Cash Runway Into 2030
As of June 30, 2026, the company had cash, cash equivalents, and marketable securities totaling $895.2 million, which is expected to provide cash runway into 2030.
William Blair notes that ELVN-001 has potential to address patients across the treatment continuum, with an initial focus on the growing population of late-line patients followed by expansion into earlier lines of therapy.
William Blair Sees $8.8 Billion CML Market
Analyst Andy Hsieh wrote that despite multiple approved tyrosine kinase inhibitors for CML on the market, unmet need remains as a growing pool of late-line patients seek more effective, tolerable, and convenient treatment options.
William Blair estimates the current total addressable market for CML is roughly $8.8 billion, including $5.6 billion from the U.S.
The analyst also noted that the 48% MMR achievement rate at the planned Phase 3 dose level is two times the rate demonstrated by current market leader Novartis AG’s (NYSE:NVS) Scemblix (asciminib).
Moreover, ELVN-001 is the only TKI to have demonstrated activity against treatment-emergent resistance mutations to Scemblix.
The analyst said ELVN-001 could disrupt the chronic myeloid leukemia treatment landscape, citing its differentiated profile and potential to serve patients across multiple lines of therapy.
