Structure Therapeutics Inc. (NASDAQ:GPCR) on Tuesday announced encouraging clinical trial findings for two of its main oral drug candidates targeting metabolic conditions and chronic weight management.
Topline results from an early-stage trial of ACCG-2671 and 72-week extension data for aleniglipron demonstrated strong weight loss effects and favorable tolerability.
Positive Early Data For ACCG-2671
The Phase 1/2a single ascending dose study of ACCG-2671 evaluated 31 healthy, non-obese participants across doses ranging from 1 mg to 10 mg.
The candidate exhibited rapid absorption and a terminal half-life of roughly six days, supporting daily or weekly administration.
Participants receiving a single 10 mg dose achieved a 3.3% mean body weight reduction at Day 24.
Furthermore, the treatment reduced bone resorption biomarker CTX-1 by approximately 60% on Day 2.
The trial recorded no serious adverse events or liver enzyme elevations, though dose-related gastrointestinal side effects appeared at higher doses.
Following these outcomes, the company started a multiple ascending dose trial, with data expected in the first half of 2027.
Extended Weight Reductions With Aleniglipron
In the ACCESS open-label extension study, aleniglipron demonstrated sustained weight loss through 72 weeks in adults continuing daily oral treatment.
Patients receiving the 180 mg maximum dose reached up to 16.2% body weight loss without reaching a plateau.
Starting with a lower 2.5 mg dose enhanced gastrointestinal tolerability, and fewer than 5% of participants stopped treatment due to adverse events.
Ongoing Milestones And Phase 3 Program
These findings reinforce the ongoing Phase 3 ACCOMPLISH program evaluating aleniglipron, which initiated in August 2026 with topline data anticipated in the second half of 2028.
Additionally, the company expects to report three distinct trial readouts in the fourth quarter of 2026 focusing on body composition, type 2 diabetes, and transitioning patients from injectable GLP-1 medicines to oral aleniglipron.
GPCR Price Action: Structure Therapeutics shares were down 8.45% at $43.34 at the time of publication on Tuesday, according to Benzinga Pro data.
