Silexion Therapeutics Corp. (NASDAQ:SLXN) shares rose 60.99% to $0.40 in after-hours trading Monday after the company reported positive preclinical findings for its lead candidate, SIL204.

Silexion Therapeutics is a clinical-stage biotechnology company developing RNA interference therapies for KRAS-driven cancers.

Preclinical Findings

The company said a translational study found that SIL204 uses naturally occurring circulating lipoproteins to achieve cellular internalization, driving dose-dependent knockdown of mutant KRAS oncogene expression in cancer cells, according to a press release.

In the study, human KRAS-driven cancer cells treated with SIL204 in a carrier-free assay without synthetic transfection reagents showed dose-dependent gene silencing in the presence of standard physiological serum lipids. Depletion of serum lipids led to a statistically significant reduction in knockdown efficiency (P < 0.001).

The company said adding exogenous human low-density lipoprotein did not change gene-silencing potency compared with complete serum alone, indicating that endogenous lipoprotein levels are sufficient for cellular uptake without artificial carrier vehicles.

“These findings represent an essential mechanistic milestone in our translational characterization of SIL204,” said Dr. Mitchell Shirvan, Chief Scientific and Development Officer of Silexion Therapeutics.

Ilan Hadar, Chairman and CEO of Silexion Therapeutics, said the findings “reinforce our broader clinical development strategy,” adding that they provide “a strong mechanistic foundation for systemic subcutaneous delivery” as the company continues site initiation and patient screening in its Phase 2/3 program.

The findings come as Silexion continues clinical trial site initiation at Tel Aviv Sourasky Medical Center and regulatory clearances in Germany.
