The U.S. Food and Drug Administration (FDA) on Wednesday approved Pfizer Inc.’s (NYSE:PFE) TUKYSA (tucatinib) in combination with trastuzumab and pertuzumab.

This regimen serves as a maintenance therapy for adults battling unresectable locally advanced or metastatic HER2-positive breast cancer following their initial induction treatment.

Promising Clinical Trial Outcomes

This regulatory decision stems from the robust HER2CLIMB-05 Phase 3 trial. Patients receiving the TUKYSA combination experienced a 35.9% reduction in the risk of disease progression or death compared to those taking a placebo alongside trastuzumab and pertuzumab.

The median progression-free survival reached 24.9 months for the TUKYSA arm, outperforming the 16.3 months observed in the placebo group. This provides patients with an additional 8.6 months of life without their cancer advancing.

Expanding Front-Line Treatment Options

TUKYSA already holds a Category 1 designation from the National Comprehensive Cancer Network as a second-line or later therapy for HER2-positive metastatic breast cancer in the U.S.

This new approval elevates the drug to the front-line setting. It provides a valuable chemotherapy-free maintenance alternative, helping physicians tailor individual treatment strategies while successfully delaying disease progression.
