The U.S. Food and Drug Administration (FDA) on Tuesday removed a clinical hold on the investigational new drug application for Filana Therapeutics, Inc. (NASDAQ:FLNA).
The regulatory decision clears the path for the biotechnology company to launch a Phase 2a proof-of-concept clinical trial evaluating simufilam as a treatment for Tuberous Sclerosis Complex (TSC)-related epilepsy.
The company anticipates screening the first patients for this trial by the first quarter of 2027.
In December 2025, the FDA issued a clinical hold on the trial, seeking additional information, including additional preclinical data, and to modify the protocol design.
Upcoming Phase 2a Trial Details
Filana designed the Phase 2a trial as a 16-week, double-blind, randomized, and multicenter study. Researchers will test two doses of simufilam.
The company plans to enroll 40 participants between the ages of 12 and 55. Participants who successfully finish the initial double-blind treatment period can enter a 48-week extension phase.
Medical teams will evaluate safety, tolerability, and pharmacokinetics. They will also measure specific seizure metrics, such as frequency, intensity, duration, nighttime occurrences, and impacts on sleep.
The Science Behind Simufilam
Simufilam is an oral small molecule designed to modulate the filamin A protein. Two separate preclinical mouse models back the clinical testing of this drug for TSC-related epilepsy.
Furthermore, independent research conducted alongside the TSC Alliance Preclinical Consortium utilized a recognized TSC-knockout mouse model.
This research revealed that simufilam reduced the progression of seizure activity in a dose-dependent fashion.
To support this ongoing work, the company secured an exclusive worldwide intellectual property license from Yale University for applying simufilam to TSC-related epilepsy.
In March, Cassava Sciences rebranded as Filana Therapeutics to reflect its strategic focus on developing novel medicines to modulate the filamin A protein for central nervous system disorders.
